Pantethine is a stable and bioavailable form of vitamin B5 (pantothenic acid), well-recognized for its favourable effects on markers of cardiovascular disease.

After initial conversion to pantetheine (two molecules of pantethine linked by a disulfide bridge) by intestinal bacteria, it is ultimately used for the biosynthesis of coenzyme A (CoA). CoA is a carrier of acetyl groups critical to multiple metabolic processes within the cell, including the Krebs (citric acid) cycle, as well as fatty acid and phospholipid metabolism.

Pantethine is commonly referred to as the “anti-stress vitamin” because of its broad utility for a wide range of processes, including energy production, cellular signalling, and the regulation of inflammation and immune function.

Case-control analysis indicates that higher pantothenic acid levels are associated with a lower risk of coronary heart disease, likely mediated via several mechanisms.

Controlled trials with participants at moderate risk of cardiovascular disease, with no inhibition of CoQ10 synthesis, showed that pantethine specifically improves dyslipidemia, including reducing non-HDL-C, triglycerides, and resulting in as much as an 11% decrease in LDL-C (vs. 3% increase with the placebo).6–8 Additionally, pantethine provides protection against LDL-C peroxidation, an independent risk factor for atherosclerotic cardiovascular disease. A vitamin B5 deficiency has been associated with hypertension, likely because of its requirement for adrenal function.
